See HELP CLASS. Beware, though, Eagle sometimes doesn't do the Right Thing, so check the results carefully. I had one design that had some high-voltage traces, so I created a couple of classes with larger clearance values. However, I found that in a few instances, Eagle routed a low-voltage net too close to a high-voltage net because it used the smaller clearance value instead of the worst-case (largest) value among the two nets. I had to fix this manually.
