What OS are you running on your P4? If it's NT (including W2K, XP) you gotta smack it upside the head to get it to let you at the ports. What version of Windows are you running this under? Have you tried just telling MPLab to use it as a PICStart+? Also, do make sure that you have it connected to the serial port you say you do, and that you have power to the unit.
